Анотація: This handbook provides comprehensive reference information on the efficient production of secondary metabolites from plants by transgenesis and other genetic manipulation strategies. It reviews and summarizes selected important case studies in genetic methods applied to enhance the production of a given metabolite or a group of related or derived compounds. Readers will find reference information on a multitude of techniques and methods, including traditional breeding and screening; over-expression of genes encoding key enzymes; functional genomics approaches; metabolic engineering of the relevant biosynthetic pathways; indirect genetic approaches to improve metabolite production, including Agrobacterium mediated transformations. Furthermore, combinatorial biochemistry approaches to engineer secondary metabolic pathways are summarized, which can offer access to new structures or to the design of novel compounds. Since many commercially valuable substances are still extracted from plants, being largely inaccessible to efficient modern laboratory synthesis methods, this book provides a valuable resource of information for biotechnological approaches that can help to find alternative and more efficient methods for the production of natural secondary metabolites. Thus adjusted production methods, with the help of tailored plant systems, can potentially help to release the stress on plants, which are currently suffering from extensive human harvesting, and to conserve global biodiversity. Readers will find comprehensive reference information on plant genetic manipulation toward more efficient synthesis, accumulation and production of target secondary metabolites. The handbook will appeal to researchers and professionals, but also graduate students and scholars working in the fields of biotechnology, genetic engineering, medicinal plant research, pharmacy, and phytochemistry.

Анотація: The series Topics in Current Chemistry Collections presents critical reviews from the journal Topics in Current Chemistry organized in topical volumes. The scope of coverage is all areas of chemical science including the interfaces with related disciplines such as biology, medicine and materials science. The goal of each thematic volume is to give the non-specialist reader, whether in academia or industry, a comprehensive insight into an area where new research is emerging which is of interest to a larger scientific audience. Each review within the volume critically surveys one aspect of that topic and places it within the context of the volume as a whole. The most significant developments of the last 5 to 10 years are presented using selected examples to illustrate the principles discussed. The coverage is not intended to be an exhaustive summary of the field or include large quantities of data, but should rather be conceptual, concentrating on the methodological thinking that will allow the non-specialist reader to understand the information presented. Contributions also offer an outlook on potential future developments in the field.

Анотація: In this thesis, applications of aminoacylation ribozymes named flexizymes are described. Flexizymes have the following unique characteristics: (i) substrate RNA is recognized by two consecutive base pairs between the 3'-end of substrate RNA and the 3'-end of the flexizyme; (ii) these base pairs can be substituted with other base pairs; and (iii) various activated amino acids can be used as substrates including both canonical and noncanonical amino acids. This flexible aminoacylation of RNAs by flexizymes was used to label endogenous tRNAs to be removed, and in vitro selection using the tRNA-depleted library enabled the discovery of the novel interaction between the microRNA precursor and metabolites. Flexizymes are also used to prepare various aminoacyl-tRNAs bearing mutations at the 3'-end to engineer the translation machinery and to develop the orthogonal translation machinery. The first part of the research demonstrated that SELEX is appropriate for discovering the interaction between small RNA and ligands, and suggested that more RNA motif binding to small molecules exists in small RNAs. The second part opened a door to new opportunities for in vitro synthetic biology involving the engineering of the genetic codes and translation machineries. This research also indicated the great potential of aminoacylation by flexizymes to be applied in various fields of RNA research, which is beneficial for RNA researchers.

   Gene Function.

Анотація: This volume focuses on the modulation of biological membranes by specific biophysical properties. The readers are introduced to emerging biophysical approaches that mimick specific states (like membrane lipid asymmetry, membrane curvature, lipid flip-flop, lipid phase separation) that are relevant to the functioning of biological membranes. The first chapter describes innovative methods to mimic the prevailing asymmetry in biological membranes by forming asymmetrical membranes made of monolayers with different compositions. One of the chapters illustrates how physical parameters, like curvature and elasticity, can affect and modulate the interactions between lipids and proteins. This volume also describes the sensitivity of certain ion channels to mechanical forces and it presents an analysis of how cell shape is determined by both the cytoskeleton and the lipid domains in the membrane. The last chapter provides evidence that liposomes can be used as a minimal cellular model to reconstitute processes related to the origin of life. Each topic covered in this volume is presented by leading experts in the field who are able to present clear, authoritative and up-to-date reviews. The novelty of the methods proposed and their potential for a deeper molecular description of membrane functioning are particularly relevant experts in the areas of biochemistry, biophysics and cell biology, while also presenting clear and thorough introductions, making the material suitable for students in these fields as well.

Анотація: This is the first comprehensive reference compilation on the substance class of glucosinolates. This handbook introduces the reader to the sulfur-containing glucosinolates (S-glucosides), a class of secondary metabolites of almost all plants of the order Capparales, in particular in the family Brassicaceae (e.g. broccoli and other cabbages), derived from glucose and an amino acid. The book illustrates the natural variety of glucosinolate structures, mainly derived from the precursor amino acid. Chapters describe the resulting rich bioactivity of the glucosides, for example as anti-cancer agents, insecticides, nematicides, fungicides, their potential phytotoxic effects, antimicrobial activity and their possible role in neurodegenerative diseases and human health. Different methods for the extraction, characterization, quantification and processing of the glucosinolates are introduced, and potential applications are discussed. The fate of glucosinolates during food processing is also briefly addressed. This handbook is written by leading experts and structured in different sections addressing the natural occurrence of glucosinolates, their (bio-)synthesis, bioactivity, food processing of glucosinolate-containing vegetables, health and disease-related aspects, biotechnology, and methods applied in glucosinolate-research. It is thus a rich reference source for every reader working in the field, from chemists and biotechnologists, to life scientists, pharmacists and medical scientists.

Анотація: This handbook compiles authoritative information about fungal metabolites and their chemistry and biotechnology. The first in the reference work series “Phytochemicals”, and written by a team of international expert authors, this book provides reference information ranging from the description of fungal natural products, over their use e.g. as anticancer agents, to microbial synthesis, even spanning to the production of secondary metabolites on industrial scale. On the other hand it also describes global health issues related to aflatoxin production in foods and agriculture, including perspectives for detoxification. The handbook characterizes different compound classes derived from fungal secondary metabolites, like ergot alkaloids and aflatoxins. The discussion puts a special emphasis on how potentially useful compounds can be obtained and what applications they can find, on the one hand, and how potential dangers can be encountered on the other hand. The comprehensive chapters in this handbook will thus appeal to readers from diverse backgrounds in chemistry, biology, life sciences, and even medicine, who are working or planning to work with fungal (secondary) metabolites and their application.
