Анотація: In the last decade, immunotherapy has revolutionized the practice of medicine in the field of oncology. This book, Immunotherapy, highlights the immunotherapeutic approaches currently in use and strategies to overcome the associated challenges. As comprehensive knowledge of basic immunology in the context of tumor biology is required to move further along the line of development in translational science, this book provides an overview of the cellular components of the immune system that interact with each other to provide a successful immune response. Immunotherapy offers promising antitumor activity across multiple cancer types as it does not attack the tumor but primarily revitalizes a suppressed immune system. It is beyond the scope of this book to discuss the immunotherapeutic approaches across the tumor board. However, the book features the role of immune-oncology in acute myeloid leukemia, non-small cell lung cancer, and sarcomas to showcase the recent advances in the management of cancer broadly classified as hematological malignancies, solid tumors, and tumors arising in the connective tissue. The book also discusses alternative formulations and combinatorial approaches to enhance the therapeutic index of these agents. As immunotherapeutic agents differ significantly from chemotherapeutic agents in response patterns and toxicity profiles, evaluating their safety and efficacy in clinical trials remains a challenge. In this book, we discuss the use of a variety of traditional and new immunotherapy criteria to evaluate response, their inadequacies, and clinical implications. The book also outlines how recent advances in medical imaging technology have the potential to provide high-dimensional data to improve decision support and individualize treatment selection and monitoring. Importantly, the book attempts to increase the awareness to immune related adverse events and the need for aggressive and judicious management of toxicities to improve treatment outcomes in patients on immunotherapy In short, this book provides a broad understanding of immunotherapy and ways to leverage the immune system to make inroads in the fight against cancer.

Анотація: This book offers a state-of-the-art report on recent discoveries concerning viral, bacterial, and parasite infectious cancers. Cancer is one of the most common causes of death and diseases in human populations, and 15%-25% of human cancers in worldwide are considered to result from chronic infection by pathogens. Most oncology textbooks address genetic mutation, but not infectious agents such as viruses, bacteria and parasites. As such this book stimulates further research in the new area between cancers and chronic infection, and discusses the epidemiology and molecular biology of infectious causes of cancers. It also explores the prevention and treatment of infection-related cancers, and brings pathogenic research to the forefront in the never-ending endeavor to understand how pathogens maneuver and negotiate in a complex environment, including the micro/macro- environment of the human host. Further, it highlights the urgent need for a concerted program to develop vaccines and other diagnosis and interventions that will eventually help prevent and treat infectious cancers, and decrease their burden on human populations. It offers graduate students and researchers a comprehensive overview of the infectious causes of cancers.

Анотація: This edited volume is about novel targeted therapies for the treatment of advanced neoplasms of the head and neck (HNSCC) with particular reference to oral squamous cell carcinomas and covers specific fields of cancer genetics and genomics for personalized medicine. The landmark studies from the HNSCC Cancer Genome Anatomy (TCGA) and the cancer Genome Atlas 2015 have revealed the multiplicity and diversity of genetic alterations in HNSCC. This remarkable body of information has elucidated novel druggable targets (eg. EGFR, mTOR, p53, survivin and aberrant microRNA) for therapeutic interventions in head and neck cancer. Readers will discover the importance of the biology and pathology of squamous cell carcinomas, the role of cancer stem cells and other factors that contribute to resistance to curative therapies such as hypoxia. We present an in-depth analysis of the HNSCC oncogenome describing the distinct molecular alterations in squamous cell carcinomas, which participate in specific molecular pathways whose dysregulation may contribute to most HNSCC cases. Final chapters of the book elucidate how these important cancer-driving molecular events and molecular pathways could be targeted to treat head and neck cancers and their therapeutic applications for targeted therapies based on clinical trials reported worldwide. This book is aimed at a broad audience, including scientists, physicians, surgeons, and post graduate students engaged in oncology and provide the readers with an up-to-date, interdisciplinary knowledge and tools to better understand the evolving areas of targeted gene therapy and the key concept of personalized medicine in head and neck cancer.

Анотація: This volume provides the current understanding of death receptor's/TLR3 signaling regulation in cancer. Death receptors, including TRAIL-R1, TRAIL-R2, Fas and TNF-RI, owing to their ability to trigger apoptosis and to contribute to the elimination of cancer cells by the immune system have been considered, to variable extent, as important therapeutic targets for cancer therapy. But an increasing body of evidence suggests that some of these receptors may also contribute to tumorigenesis, or that new players such as TLR3 may be targeted for cancer therapy due to their ability to behave like death receptors.

Анотація: This astute volume brings together the latest expert research on adamantinomatous craniopharyngiomas (ACPs). ACPs are histologically benign but clinically aggressive tumors exhibiting a high propensity for local invasion into the hypothalamus, optic and vascular structures. These tumors, as well as the current treatments, may result in pan-hypopituitarism, diabetes insipidus, morbid obesity followed by type II diabetes mellitus, blindness, as well as serious behavioral and psychosocial impairments. Exploring in detail advances in both the understanding of tumor biology as well as clinical advances in patient management are explored in detail, this book will also look towards potential new treatment approaches. Basic Research and Clinical Aspects of Adamantinomatous Craniopharyngioma is the first book compiling all current research on ACPs. Mouse and human studies have unequivocally demonstrated that mutations in CTNNB1 encoding -catenin underlie the etiology of the majority, if not all ACP tumors. Genetic studies in mice have shown that ACPs are tumors of the pituitary gland and not of the hypothalamus as previously thought, and are derived from Rathke’s pouch precursors. In addition, a role for tissue-specific adult pituitary stem cells has been revealed as causative of ACP. Together, these studies have provided novel insights into the molecular and cellular etiology as well as the pathogenesis of human ACP. Finally, this volume covers new treatment approaches that have been shown to be effective both in reducing ACP burden as well as reducing the morbidity associated with therapy. .

Анотація: Using a multidisciplinary approach, this book describes the biochemical mechanisms associated with dysregulation of proteases and the resulting pathophysiological consequences. It highlights the role and regulation of different types of proteases as well as their synthetic and endogenous inhibitors. The role of proteases was initially thought to be limited to general metabolic digestion. However, we now know that the role of protein breakdown is much more complex, and proteases have multiple functions: they are coupled to turnover and can affect protein composition, function and synthesis. In addition to eliminating abnormal proteins, breakdown has many modulatory functions, including activating and inactivating enzymes, modulating membrane function, altering receptor channel properties, affecting transcription and cell cycles and forming active peptides. The ubiquity of proteases in nature makes them an important target for drug development. This in-depth, comprehensive is a valuable resource for researchers involved in identifying new targets for drug development. With its multidisciplinary scope, it bridges the gap between fundamental and translational research in the biomedical and pharmaceutical industries, making it thought-provoking reading for scientists in the field.
